We can't always predict how each area of the field will respond to applied fertilizer due to soil variability. Nutrient levels and availability can be affected in part by soil properties and weather conditions. Because of this, we often see nutrient deficiencies.
We couple soil sampling along with tissue sampling to observe how successful the plant uptake of nutrients is, compared to what is available in the soil to see why a nutrient may be limited. Using both soil and tissue sampling, we can see what nutrient is limited, and whether or not an in-season application will benefit.
Tissue sampling can also be taken to aid in disease identification.
